HBC ligands
HBC is nonfluorescent in solution, but emits strong fluorescence upon forming tight complex with Pepper RNA aptamer.
HBC fluorescence can be activated thousands fold by Pepper, enabling visualization of RNAs tagged with Pepper with the best signal/background contrast currently available.
There are 8 kinds of HBC ligands with different colors in HBC series fluorescent dyes. They cover a wide range of emission wavelengths, named after the maximum emission wavelength, including HBC485, HBC497, HBC508, HBC514, HBC525, HBC530, HBC599, and HBC620.
Supplied as solid formulation. Reconstitute with DMSO (60 ul for S size, or 200 ul for L size as labeled on the vial) to make 1000X stock solutions.
We suggest users start with HBC530, the orginal Pepper ligand, and HEK293T cells expressing Pepper or Pepper tandem arrays as positive control, before test your own cells and/or RNAs.
As far as we know, cells of different origin expressing Pepper alone or Pepper array show bright fluorescence and low background when stained with HBC530.
After the first success, users may proceed to test the performance of other HBC ligands on your RNA, cells and instruments.
Important Notice: Customers should include at least one positive control plasmids (PET2808MCS0 for E. coli or PAPU608MCS0 for mammalian cells) in their first order along with their choice of HBC dyes; alternatively, customers may order the Pepper Fluorescent RNA starter kits (Cat# PET2808KIT or PAPU608KIT).
